چکیده

Angiotensin II (Ang II), an important component of the renin–angiotensin system (RAS), plays a critical role in pathogenesis cardiovascular disorders. In addition, human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) have been considered as promising platform for studying personalized medicine heart diseases. However, whether Ang can induce apoptosis hiPSC-CMs is not known. this study, we treated with different concentrations [0 nM (vehicle control), 1 nM, 10 100 ?M, and mM] various time periods (24 h, 48 6 days, days) analyzed viability hiPSC-CMs. We found that treatment mM days reduced by 41% (p = 2.073E-08) increased 2.74-fold, compared to control group 6.248E-12). MYOG, which encodes muscle-specific transcription factor myogenin, was also identified apoptosis-suppressor gene II-treated Ectopic MYOG expression decreased Further analysis RNA sequencing (RNA-seq) data illustrated myogenin ameliorated II-induced downregulating proinflammatory genes. Our findings suggest induces attenuates apoptosis.

Calcium Handling in Human Induced Pluripotent Stem Cell Derived Cardiomyocytes

BACKGROUND The ability to establish human induced pluripotent stem cells (hiPSCs) by reprogramming of adult fibroblasts and to coax their differentiation into cardiomyocytes opens unique opportunities for cardiovascular regenerative and personalized medicine. In the current study, we investigated the Ca(2+)-handling properties of hiPSCs derived-cardiomyocytes (hiPSC-CMs). METHODOLOGY/PRINCIPA...

Fingolimod Enhances Oligodendrocyte Differentiation of Transplanted Human Induced Pluripotent Stem Cell-Derived Neural Progenitors

Multiple sclerosis (MS) is an autoimmune disease which affects myelin in the central nervous system (CNS) and leads to serious disability. Currently available treatments for MS mainly suppress the immune system. Regenerative medicine-based approaches attempt to increase myelin repair by targeting endogenous progenitors or transplanting stem cells or their derivatives. Fingolimod exerts anti-inf...

In vivo differentiation of induced pluripotent stem cell-derived cardiomyocytes.

BACKGROUND Induced pluripotent stem cells (iPSCs) hold promise for a new era in treating heart failure. However, the functional microstructure of iPSC-derived cardiomyocytes (iPSC-CMs) and their ability to attach to the extracellular matrix of the recipient myocardium require further elucidation. Thus, we analyzed the functional microstructure and adhesion molecules of iPSC-CM. METHODS AND RE...

Functional cardiomyocytes derived from human induced pluripotent stem cells.

Human induced pluripotent stem (iPS) cells hold great promise for cardiovascular research and therapeutic applications, but the ability of human iPS cells to differentiate into functional cardiomyocytes has not yet been demonstrated. The aim of this study was to characterize the cardiac differentiation potential of human iPS cells generated using OCT4, SOX2, NANOG, and LIN28 transgenes compared...
